Synthesis of some substituted pyrimidines as possible anti-cancer agents

Fourteen 2-phenyl/2-methyl-4-hydroxy-5-(.beta.-hydroxyethyl)-6-alkyl substituted pyrimidines were synthesized by condensation of .varies. -acyl- .gamma. -butyrolactone with benzamidine hydrochloride or acetamidine hydrochloride in presence of sodium methoxide. Anti-cancer evaluation of some of the intermediary lactones and 2-phenyl-6-alkyl substituted pyrimidines was carried out against transplantable tumors. Amongst these, 4 compounds from 2-phenyl series exhibited anti-cancer activity against Dalton's lymphoma (ascites); while none of them showed promising anti-cancer activity against standard P388 lymphocytic leukemia.
