In 2 experiments, 10-week-old pups were twice vaccinated by subcutaneous inoculations of X-irradiated Ancylostoma caninum larvae. The immunity of the vaccinated pups and of similar but unvaccinated control pups was then challenged either by exposure to an environment that was contaminated with hookworm larvae or by subcutaneous inoculation of larvae. Hookworm burdens from the challenge infections were reduced by 88 to 95% in the vaccinated pups compared with the control pups and the degree of protection in the vaccinated pups was the same, regardless of the mode of infection. All vaccinated pups survived challenge in good health, whereas the control pups were severely affected clinically and in the 2nd experiment, 7 of the 10 control pups died of severe acute ancylostomiasis.
