The chemical and antigenic nature of the two fractions of exoantigen, EA1 and E2, of Leishmania donovani have been studied. The fractions contain protein and glycoprotein as revealed by polyacrylamide gel electrophoresis. Sugar analysis showed that both fractions contained arabinose, galactose, glucose and mannose but in different molar ratios. The exoantigens gave precipitin reaction with Con-A and PHA. In counter current immunoelectrophoresis, the fractions showed multiple precipitin bands against antipromastigote sera and kala-azar paient sera. Positive reactions were also observed when the two fractions were treated with trypsin. The antigen moiety of the fractions was found to be present in the aqueous layer after hot phenol treatment. The fractions gave two peaks in rocket immunoelectrophoresis against the antisera raised with EA.
