Cell cycle phases of L. donovani promastigotes, 2S strain, both from stock cultures (SC) and recent isolates from hamster spleen (RI) were determined by the Howard and Pelc method. Phases were determined for organisms incubated at 25, 35 and 37 degrees C. RI had a longer S phase than SC at 25 degrees C but at 35 degrees C the S phase of SC was lengthened to equal that of RI. S phases of both sources were shortened by exposure to 37 degrees C. Alterations in cycle phases of the kinetoplast paralleled those of the nucleus. Variations in S produced alterations in the G1 rather than G2 or division phase. The results indicated that recently isolated parasites are more resistant to elevated temperature than are stock cultures. The nucleus and kinetoplast vary similarly in their response to hamsters passage and elevated temperature, and stock cultures undergo changes in cell cycle phases under exposure to elevated temperature making them similar to recently isolated forms.
