A simple method for cryopreservation of Leishmania donovani promastigotes

A simple and inexpensive method for short-term cryopreservation of different strains of Leishmania donovani is described. Promastigotes of L. donovani were cultured in Novy-MacNeal-Nicolle (NNN) medium and Tobie's medium and then culture tubes were kept without preservative at -80 degrees C. They were removed for examination after 9, 12, 15, 20, 23, 26, 29, 32, 42, 56, 65 and 126 days. They were kept at room temperature for 30 minutes before examination. Living promastigotes were demonstrated in each case and 2 successful subcultures were made from each cryopreserved culture.
