Novobiocin was administered to healthy men to ascertain its pattern of absorption, distribution, and excretion. Serum levels, promptly achieved, were similar by either the intramuscular or intravenous route. On oral administration in divided doses, higher levels were demonstrated when the initial dose was taken on an empty stomach. On continued oral therapy, the build-up in serum concentration was not increased after the third dose. Novobiocin was not recovered from the spinal fluid, although antibiotic activity was detected in blood withdrawn simultaneously with the tap. High urinary levels of novobiocin were assayed. Recovery of the antibiotic ranged from 0.3 to 14.7 per cent. In this limited study, the antibiotic was well tolerated regardless of the route of administration. No toxic or side effects were observed.
