Novobiocin; serum concentrations and urinary excretion following oral administration in man

1. Novobiocin has been administered orally to 60 men in both single and multiple doses at various dosage levels. Complete records of serum concentrations obtained after each dose have been presented. The results agree with and augment those obtained by other investigators of this antibiotic. 2. As compared with other antibiotics novobiocin produces extremely high blood concentrations. Following single 0.25 Gm. doses, the average peak serum level was 10.9 Ag./m1.; following 0.5 Gm. doses, it was 18.8/lg./mt.; following 1.0 Gm. dose, it was 42.4 μg./ml.; and following 2.0 Gm. doses, it was 67.7 μg./ml. 3. On the basis of the sensitivity of susceptible organisms and the blood levels obtained with this drug, it would appear that doses of 0.5 Gm. twice daily would be quite sufficient therapeutically. 4. The urinary excretion of novobiocin in man is quite low and amounts to 2.7 to 3.3 per cent of the dose administered.
