Isolation and amino acid composition of alpha-amylase from human granulocytes

A purified preparation of human granulocyte alpha-amylase was obtained by means of molecular-sieve chromatography and ion-exchange chromatography. The alpha-amylase from the granulocytes differs in its amino acid composition from alpha-amylase from the salivary glands and pancreas. It contains a greater number of basic amino acids than the enzymes from the salivary glands and pancreas. The ratio of polar to non-polar amino acids is higher in the enzyme from the granulocytes-1.76, as compared with pancreatic alpha-amylase-1.58, and salivary gland alpha-amylase-1/57 (4, 10). The differences in the amino acid composition may be responsible for the differences in the isoelectric points and in the molecular weight between this amylase and those from the pancreas and salivary glands (11, 19).
