1. Novobiocin was administered by the usual routes to mice, rats, guinea pigs, cats, rabbits, and dogs in acute, subacute, and chronic toxicity studies. 2. Acute toxicity studies indicated that novobiocin had a moderate toxicity for mice. Guinea pigs were more susceptible. 3. The short term intravenous study in dogs indicated that the liver and kidney were apparently affected at high doses (100 ung./Kg./day) and that a concentration of 130 mg./ml. was irritating to the veins. 4. Long term oral toxicity studies in rats and dogs did not, disclose any significant pattern of toxicity for novobiocin. 5. A. long term subcutaneous toxicity study in cats showed no effect on the functioning of the eighth cranial nerve (hearing and equilibrium) lior arty systemic toxicity due to novobiocin. The repeated daily subcutaneous injections for 70 days caused considerable local tissue injury. 6. Novobiocin was moderately irritating to rabbit skeletal muscle. 7. Pharmacodynamic studies did not disclose any alarming properties of novobiocin.
