A method of processing to infer and achieve the required number of motile spermatozoa post thaw, per dose of frozen semen

Samples from 217 bull ejaculates being routinely processed were subjected to a test freeze (2-h equilibration) to predict post-thawing motility. Since each semen dose was diluted to contain 30X106 spermatozoa, a post-thawing survival rate of more than or equal to 35% gave 10X106 motile spermatozoa/dose, which was considered the optimum. In order to use the numbers of spermatozoa in excess of this requirement, 104 samples showing a post-thawing motility of more than or equal to 40% were taken for the experiment, and 77 samples were diluted further. Post-thawing motility after the further dilution was 54.4% v 54.6 for samples not diluted further. Semen doses subjected to the further dilution contained, on average, 11X106 motile spermatozoa. Routine adoption of this method increased the number of doses produced by 17%.
