Production of fatty liver with dietary ethanol despite orotic acid supplementation

Young male Wistar rats weighing 50 g were given liquid diets in which casein supplied 18, fat (olive oil:maize oil: cod liver oil 12:15:1.5) 35 and carbohydrate 47% of energy intake or in which, at the expense of carbohydrate, ethanol or ethanol and orotate supplied 36% of energy; concentration of orotate was 0.3 and 0.8% of the nonaqueous portion of the diet. After 28 days on the liquid diets the rats were killed and livers were removed. Mean hepatic triglyceride concentration was significantly greater in the groups given ethanol or ethanol and orotate than in those not given ethanol, but there was no significant difference in triglyceride concentration or in the development of steatosis between groups given ethanol alone or ethanol and orotate.
