Primary selection of milk-clotting actinomycetes on solid laboratory media

A method was developed for the rapid selection of milk-clotting actinomycetes on a dense 1% casein substrate. In experiments with 7 mesophilic strains of actinomycetes, at substrate pH 5.2 and temp. of 37 and 55 deg C, acid proteases with milk-clotting activity were found. On a substrate of pH 8.5, treated with 5% TCA to precipitate the non-hydrolysed casein, proteolytic enzymes of the trypsin type were detected.
