Distribution pattern of milk-clotting and proteolytic activities in some fungi

Extra- and intra-cellular milk-clotting and proteolytic activities were measured in 10 fungi (Aspergillus flavus, 2 strains of Asp. niger, Byssochlamys fulva, Chaetomium globosum, Fusarium moniliforme, Gliocladium roseum, Mucor hiemalis, Penicillium chrysogenum and P. citrinum) grown at 30 deg C in 2% maize steep liquor + 2% lactose, with or without shaking for periods of 3, 7 or 10 days. The milk-clotting activity of the culture filtrates was highest with P. citrinum incubated without shaking. Except with the Aspergilli, the extra-cellular milk-clotting activity was lower in the shaken cultures. Cell-free extracts of the fungi (incubated with or without shaking) had low milk-clotting activity but contained significant amounts of proteolytic activity. It is concluded that the intracellular and extra-cellular proteolytic activities are different, and that probably the milk-clotting and proteolytic enzymes are also different.
