Analysis of anisodine and its metabolites in rat plasma by liquid chromatography-tandem mass spectrometry

Aim: To analyse the contents of anisodine (a constituent of Anisodus tanguticus) and its metabolites in rat plasma after ingestion of anisodine by combining liquid chromatography and tandem mass spectrometry (LC-MSn). Methods: Plasma samples from rats taken after single oral administration of 20 mg anisodine were treated with methanol to precipitate the protein. Then, it was analysed by LC-MSn. Identification and structural elucidation of the metabolites were performed by comparing their molecular masses, retention times and full scan MSn spectra with those of the parent drug and blank plasma. Results: The parent drug and its four metabolites (norscopine, scopine, hydroxyanisodine and N-oxide anisodine) existed in the rat plasma. Conclusion: This method is sensitive, rapid, simple, and suitable for the rapid identification of the drug and its metabolites.
