Characterization of antigenic property of Toxocara canis and Toxascaris leonina adults and larvae through immunodiagnostic electrophoresis (SDS-PAGE) and western blot technique

Somatic antigens of adult and larval Toxocara canis and Toxascaris leonina, recovered from naturally-infected dogs, were analysed by SDS-PAGE. Both adult somatic antigens showed 2 closely similar bands (90.00-91.95 and 69.25-70.56 kDa) which were cross reactive with Western blot. Each parasite had characteristic bands clustered at different MWs. Toxocara canis larval antigen showed a very different antigenic profile when compared with Toxascaris leonina antigen except for one band (66.85-66.89 kDa). Using hyperimmune rabbit sera and Western blot, Toxocara canis adult antigen reacted with 4 prominent bands at 125.37, 117.73, 90.00 and 69.25 kDa; the corresponding 3 bands for Toxascaris leonina were 119.04, 91.95 and 70.56 kDa. The bands at 125.37 and 117.73 kDa can be used as a specific fingerprint for Toxocara canis adults whereas the 119.04 kDa band is specific for Toxascaris leonina adults.
