The present work deals with the effects of a) single intravenous injection of: benzene, toluene, cyclohexane and hexane; and b) repeated subcutaneous injections of the same substances, in rabbits. A rapid death of the animals can always be observed after injecting: 1) 0,15 cc/kg of benzene or xylene; 2) 0,20 cc/kg of toluene or hexane, and 3) 0,30 cc/kg of cyclohexane by the intravenous route. A striking fall of granulocytes regularly follows the repeated subcutaneous injection of 1 cc/kg of benzene, 1 cc/kg of cyclohexane and 4 cc/kg of hexane. Only benzene intoxication provoked a hypoplastic marrow. Toluene produced only a transient granulopenia, even after being injected in doses that kill the animals at the end of the second day. Benzene has been given orally to 48 patients with chronic leukaemia with a fairly good tolerance and complete or partial remissions in most cases. On the contrary, toluene was unable to provoke a leukocytic drop, a negative result in leukaemic patients in accordance with our findings in rabbits.
