Non-specific leukocytolysis in vitro as a test of comparative cytotoxicity of low molecular weight nonelectrolytes

Leukocytolytic effects of urea, glycerine and glucose on the blood of intact dogs were studied. Hypo-, iso- and hyperosmotic solutions were used in equimolar concentrations in order to standardize osmotic conditions. Leukocytolytic effects of all the three non-electrolytes had a parabolic pattern depending on the osmotic concentrations. Comparative leukocytosis degree was higher in urea than in glycerine and glucose. It was in inverse proportion to the agent molecular weights. As these differences are due to the chemical component of the effect, nonspecific in vitro leukocytolysis is considered a test for comparative cytotoxicity, as opposed to a specific (immunologic) one.
