Effects of trimethoprim-sulfamethozypyrazine combination on the reproduction in rats and rabbits

Experimental studies on Sprague-Dawley rats, were carried out in order to distinguish the different phases of the reproducing process. It was found that a trimethoprim-sulfamethoxypyrazine combination, administered at doses from 2 to 8 times higher than the maximum therapeutic dosage, caused no embryotoxic or teratogenic effects and did not inhibit fertility. At higher doses, the combination appeared to inhibit fertilization of the egg and to induce cataract in a low percentage of fetuses. In the New Zealand White rabbit, doses even 25 times higher than the therapeutic dosage, caused no significant effects. The different results obtained in two analogous animal species, have confirmed the difficulty of predicting embryotoxic effects in man.
