Ultracytochemical localization of adenylate cyclase in liver of normal and of CCl4 and CCl4 plus colchicine treated rats

Previous studies have demonstrated an adenylate cyclase (AC) system sensitive to glucagon and sodium fluoride (NaF) in the normal rat liver, localized in the sinusoidal region and/or on bile canaliculi. In the present study the cytochemical localization of AC was studied in the liver of normal and of carbon tetrachloride (CCl4) and CCl4 plus colchicine treated rats. The enzymatic activity was investigated in basal condition and after stimulation with NaF and glucagon. In all experimental conditions the AC reaction product was always present on bile canaliculi plasma membranes. On microvillar plasma membranes AC was present only after stimulation by glucagon. Moreover in this site glucagon sensitive AC was detectable only in normal liver and after treatment with CCl4+ colchicine, while it was lacking in CCl4 treated liver. These findings support previous biochemical data and confirm that colchicine plays a significant role in the protection of glucagon sensitive AC from CCl4.
