In vitro evaluation of the trypanocidal activity of 3 antimycotic agents on fresh plasma infected with T. cruzi

The incidence of positive serology for Chagas' disease is high among blood donors in Venezuela. On the other hand often there is a deficit of plasma and cryoprecipitate to treat hemophilic patients. In order to develop alternatives to this situation we evaluated if it was possible to steralize the blood from blood donors with Chagas' disease. For this study fresh normal plasma was inoculated with know concentrations of Trypanosoma cruzi (Tc) and incubated with the following antimycotics: Amphotericin B, Candicidine and Miconazol. As a primary screeing of the effect of the effect of these drugs on the parasite, tripomastigotes diluted in normal plasma were plated in flat bottom 96 wells plates (106 tripomastigotes/well) and incubated with serial dilutions of the antimycotics Amphotericin A 10 to 0.07 .mu.g/ml; Candicidine 0.6 to 0.01 .mu.g/ml; Miconazol 5 .times. 10-5 to 1 .times. 10-6 .mu.g/ml). Using loss of motility as indicator of cell death we determined the concentration of each antimycotic that induced 100% tripomastigotes mortality. In order to define more precisely the viability of parasite 2 .times. 105 and 2 .times. 106 Tc/ml were incubated, with the minimal concentration of antinycotic that induced 100% mortality, for 3 hours at 4.degree. C and inoculated in vero cells. Incubated for 14 days at 37.degree. C, processed and that presence of infected cells evaluated as more precise indicator of parasite survival. Of the three drugs tested Amphotericin B has the best tripanicidal activity. Neither Amphotericin B nor the parasites interfered with the activity of Factor VIII.
