Thin-layer and high performance liquid chromatographic study of tetramethylthionin chloride (methylene blue) used in general medicine and tetramethylthionin base for the eyes

Methylthionin chloride, used in general medicine, and tetramethylthionin base are analyzed by thin-layer chromatography with silica gel (Merck silica GF 254) and by high performance liquid chromatography, the elution solvent was the upper phase of PARTRIDGE (n-butanol acetic acid, water; 4-1-5) and a C 18 Waters .mu.-Bondapak column was used. A correlation was found between the peaks noted during HPLC and the spots observed in CCM, thus ruling out the supposition of a possible degradation of tetramethylthionin by thin-layer chromatography with silica gel.
