Complement fixation and haemagglutination reaction with methanol extracts of icterohaemorrhagiae, canicola and patoc leptospires serovars

In complement fixation (CF) tests, homologous antigens showed higher titers than heterologous antigens with antileptospira rabbit sera. Sera from blood-bank and persons infected with Chagas disease, syphillis, hepatitis and leprosy were not reactive in CF test with methanol extracts of leptospira. Antigen pools of serovars icterohaemorrhagiae, canicola and patoc, when titrated with anti-leptospira rabbit sera, showed the same titer as individual homologous antigens. Human erythrocytes (of group "O", Rh negative) were treated with chromic chloride and sensitized with methanol extracts of leptospira. In hemagglutination (HA) tests, anti-leptospira rabbit sera cross reacted with heterologous antigens. Sera from healthy individuals and patients with Chagas' disease, syphilis, leprosy and hepatitis gave nonspecific reactions at dilutions between 1;10 and 1:160. The antigens remained stable for at least 30 months. Chemical analysis of the antigens showed the presence of polysaccharides and proteins.
