Growth characteristics of colony-forming units of fibroblasts (CFU-F) in human monolayer cultures of bone marrow in various forms of hematologic diseases

The quantitative analysis of the fibroblast colonies grown in the monolayer culture revealed two types of colony-forming units of fibroblasts (CFUf) differing in their proliferative activity: those forming "large-dense" and "small-loose" colonies, comprising 15 and 85%, respectively. Such a ratio of the CFUf levels is characteristic of the normal bone marrow of acute leukemia patients during hematologic remission, and of chronic myeloleukemia patients having no blast forms in the bone marrow in the active stage of the disease. The monolayer cultures of the bone marrow of chronic leukemia patients during relapse did not demonstrate the previously detected heterogeneity of the CFUf population. The results obtained suggest certain changes in the character of the parental cells of the stromal microenvironment during relapse of acute leukemia.
