Effect of cryopreservation on recovery of megakaryocytic colony-forming units in murine bone marrow

The proliferative capacity of megakaryocytic colony-forming units (CFU-M) in cryopreserved murine bone marrow was assessed by the in vitro plasma clot method. The recovery of CFU-M, expressed as percent colonies present in the fresh bone marrow culture, was 74% on an average for the samples which had been frozen to -80.degree. C and thawed 30 min later. The viability of CFU-M, as judged by colony growth, was maintained for at least 1 yr without significant loss in frozen bone marrow stored in liquid N (-196.degree. C) while it began gradually to decline after storage for 6-8 wk in a deepfreezer (-80.degree. C). The optimal conditions for cryopreservation of CFU-M with regard to the concentration of dimethylsulfoxide, and dilution and washing techniques were nearly identical to those previously reported for granulocytic progenitor cells.
