Effect of lymphocyte-conditioned medium on human bone-marrow colony forming units-megakaryocyte originated colony formation in a patient with liver cirrhosis

To investigate the pathogenesis of thrombocytopenia seen in patients with liver cirrhosis, the effect of conditioned medium from phytohemagglutinin-P (PHA-P) stimulated human peripheral mononuclear clear (PHA-LCM) on in vitro colony forming units-megakaryocyte (CFU-Meg) originated colony formation was studied. CFU-Meg originated colonies were identified by the avidin-biotin peroxidase complex staining using human antifactor VIII associated rabbit IgG. CFU-Meg originated colonies from normal subjects were significantly reduced by the addition of the patient's PHA-LCM. In contrast, CFU-Meg originated colonies from the patients were significantly increased by the addition of the normal PHA-LCM. These results suggest that the in vitro production of some factors stimulating and/or regulating the megakaryocytopoiesis were reduced significantly in the patient's mononuclear cells. Thus mononuclear cells may contribute to the pathogenesis of thrombocytopenia seen in patients with liver cirrhosis.
