Proliferative response of T cells, B cells and macrophages during inductive phase of immunization to BCG in guinea pig

T and B cells were quantitated by using spontaneous rosette forming and surface Ig marker techniques in the lymph nodes of guinea pigs following BCG immunization. The total proliferative response was further estimated by 3H-thymidine uptake in vitro. The cellular response appeared biphasic and occurred during the 1st and 2nd wk of immunization. During the 1st wk, there was simultaneous and proportionate increase in both T and B cells associated with a very high uptake of thymidine. During the 2nd wk, macrophage accumulation and proliferation were prominent features and associated with active germinal center formation and a high 3H-thymidine uptake. Thus, in addition to the activity of T cells and macrophages, the B cells also make a significant contribution to the overall immune response.
