Effect of praziquantel on glycogen content and [1-14C] glucose uptake in Schistosoma japonicum

Glycogen content, [1-14C]glucose uptake and incorporation of [1-14C]glucose into glycogen were markedly decreased after pairs of S. japonicum were incubated in 1 or 10 mu g/ml praziquantel for 4 h. Males appeared to be affected more than females. After transfer to drug-free medium, females recovered quickly, but in males the effects caused by the higher concentration of drug were not reversible. In infected mice given praziquantel at 20 mg/kg, the glycogen content of schistosomes was markedly decreased after 2 to 4 h, but after 48 h it recovered. At doses of 100 to 300 mg/kg there was no apparent recovery of glycogen content 72 h later. Females perfused from mice 8 h after treatment with 100 to 300 mg/kg were able to recover glycogen content after in vitro incubation, but in males there was no significant recovery.
