Affinity chromatography was initially used to purify endogenously unsaturated folate binder from bovine milk [DSA 34, 2355]. Application of this technique in conjunction with techniques to dissociate the folate binder complex at acidic pH has permitted both the identification and subsequent isolation of endogenously saturated folate binders in animal tissues and fluids. This paper describes these procedures and techniques for assaying folate binding.
