Effects of papaverine on the release and metabolism of dopamine in rat striatum

Papaverine (30 and 100 .mu.M) enhanced the basal efflux of radioactive products from rat neostriatal tissue slices prelabeled with AMP (0.1 mM) and 1-methyl-3-isobutylxanthine (0.1 mM) did not affect the basal or the K+-mediated outflow of radioactive products. Pretreatment with both agents failed to modify papaverine effects on basal and K+-evoked outflow of total 3H. Apparently papaverine effects on dopamine release and metabolism are not related to cAMP accumulation. The possible relationships between an effect of a drug on storage vesicles (granular effect) and enhanced neurotransmitter release are discussed.
