A method to study bile secretion in urethane anesthetized guinea-pigs is proposed. After a ligation of the cystic duct, the common duct is catheterized in animals placed at a constant temperature and infused with sodium taurocholate. Bovine albumin (3%) added to sodium taurocholate allowed a survival up to 7 h. As control of experimental technique, a dose-response curve to secretin was determined. The dose yielding half-maximal stimulation calculated with the Lineweaver-Burk transformation was 0.26 clinical unit for volume of bile and 0.23 clinical unit for bicarbonate output. In a preliminary study, the effect of peptide extracts from porcine upper small intestine (gastrozymin) on bile secretion was tested and compared it to that of secretin; 2 gastrozymin extracts, obtained at different stages of purification, strongly stimulated bile flow and bicarbonate output; the dose-response curve was significantly different from that of secretin.
