Extracellular "in vitro" evolution of metacyclic trypomastigotes isolated from Trypanosoma cruzi cultures

Metacyclic trypomastigotes of Trypanosoma cruzi were obtained from cultures at 27 deg C and were freed from epimastigotes by exposure to fresh rat blood and then to mouse blood. They were cultivated further on LIT medium at 27 deg C and they multiplied and transformed into epimastigotes. In certain strains or conditions they also developed into short broad trypomastigotes. If the temperature was raised to 37 deg C, amastigotes appeared but further development was hindered. Apparently metacyclic trypomastigotes are not exclusively dependent on intracellular proliferation for further development.
