Trypanosoma cruzi was cultivated in vitro in a medium consisting of Bacto Liver (35g), tryptose (10g), glucose (5g), Na2HPO4 (8g), KCl (0.4g), NaCl (4g), yeast extract (3g), haemin (20g) and water (1000ml), at pH 7.8. This was sterilized at 115 deg C for 15 min. When used in a fermenter with 9 litres medium, it yielded 70g wet sediment of epimastigotes which could be used to prepare antigens for serological diagnosis. During growth the concentration of glucose and the pH fell to minimum on day 11 after which both the ammonia and the pH increased.
