Activity of Dipernoids isolated from Azorella compacta (Llareta) on Trypanosoma cruzi amastigotes

This study tested the trypanocidal activity, against amastigote forms of the SPA-14, Tulahuen and G strains and the CL Brener clone of Trypanosoma cruzi, of diterpenoids isolated from Azorella compacta, a plant used for medicinal purposes in Chile before the Spanish conquest. Of 6 compounds isolated, 2 had amastigocidal activity. Azorellanol, a diterpene isolated for the first time, showed a 50% inhibitory concentration (IC50) that varied between 60 micro M (CL Brener clone) and 84.1 micro M (SPA-14 strain), and mulin-11,13-dien-20-oic acid showed an IC50 between 41 micro M (G strain) and 87 micro M (CL Brener clone). The cytotoxicity levels of both compounds against Hela and Vero cells and J144 macrophages were generally higher than those of nifurtimox but similar to those of gentian violet; both had lower cytotoxicity against Hela cells than either nifurtimox or gentian violet.
