Tolciclate against dermatophytes

The fungistatic activity of tolciclate (O-(1,2,3,4-tetrahydro-1,4-methanonaphthalen-6-yl)-m-N-dimethylthiocarbanilate; KC 9147) was compared with that of tolnaftate against Trichophyton asteroides [T. mentagrophytes], T. mentagrophytes, Microsporum audouinii, M. canis, M. gypseum, Aspergillus niger and Epidermophyton floccosum in Sabouraud broth and in agar. The min. inhibitory concs. for tolciclate ranged from 0.012 to 0.056 mu g/ml and there was no significant difference between the 2 drugs. Activity was reduced approx X 8-10 when 10% serum was added. Tolciclate was fungicidal after some days' contact.In experimental cutaneous infections on guinea pigs, tolciclate applied topically for 10 days displayed high activity against T. mentagrophytes and M. canis. It was X 3-10 more active than tolnaflate.
