Extraction of vaccinia virus and purification of vaccinia elementary body suspension with Mafron 11

Mafron 11 (Trichloro-monofluro-methane) was used for the extraction of vaccinia virus from skin pulp and chorio-allantoic membranes (CAM). It was as effective for this purpose as Arcton 113 (Trichloro-trifluro-ethane). Sequential extraction yielded a significant increase in the viral content of the elementary body suspensions (EBS). Mafron 11 had no adverse effect on the antigenicity, reactogenicity, pathogenicity or immunogenicity. The advantage of the greater bactericidal action of Mafron 11 enabled it to be utilized for the purification of EBS. A method of achieving quick purification within an hour without loss of virus in the presence of 10% Mafron 11 and 0.125% phenol was described. Mafron 11 is superior to Arcton 113 since apart from liberating the vaccinia virus from the pulp it also purifies it.
