The chemotherapeutic activity of selected fasciolicides against immature Fasciola hepatica in mice

The efficacy of rafoxanide, oxyclozanide, nitroxynil and diamphenethide against immature stages of Fasciola hepatica in mice was investigated. The doses used were derived from preliminary toxicity studies with the drugs, and were given in diet at: rafoxanide 0.0125 and 0.025%; oxyclozanide 0.10 and 0.40%; nitroxynil and diamphenethide 0.10% for each. The mice were infected by gavage with one or 2 metacercariae, treated over different periods covering the whole of the immature phase of migration of F. hepatica and then killed either 21 or 31 days after infection. Only diamphenethide was active for all the periods tested (62 to 100% fluke reduction); the other drugs were generally ineffective over -1 to 6 days after infection, and displayed varying activity (40 to 100%) over the other periods. Flukes recovered from the treated animals were retarded in size and there were corresponding reductions in liver pathology. It is concluded that the mouse-F. hepatica system could be used for screening of potential fasciolicides.
