Toxicológical properties of diosmin and its actions on the isolated venous tissue of the dog

Toxicological properties of diosmin, a naturally occurring flavonic glycoside, and actions exerted on isolated saphenous vein strips of the dog are described. Subchronic toxicity was studied in rats; mice and minipigs were used for chronic toxicity studies. Possible teratogenic effects were searched in rats and mice. The low acute toxicity made the use of large daily doses in these experiments feasible. Toxic or teratogenic effects were not detected in any experimental animals. Diosmin markedly blocked inactivation of exogenous noradrenaline [norepinephrine, NA] and caused a slow and gradual contractile response of the oil-immersed strip, which was not attributable to release of NA. Possible mechanisms of this action, which may be causally related to clinical benefit in chronic venous insufficiency, are discussed.
