For muscarinic receptors, the years from 1951 to 1985 were the time period of emerging awareness of receptor subtypes, mainly based on functional work with newly-available selective agonists and antagonists. The rapid advances in the current decade (1985-1994) in our understanding of the molecular biology, cell biology, biochemistry and electrophysiology have greatly enhanced our understanding of the diversity (M1- M4/pharmacologically-defined receptors versus m 1 -m5/cloned receptor genes) and mechanism of action of muscarinic receptors, and their role in clinical medicine. One can expect for the next decade (1995-2004) that when novel subtype-selective muscarinic agonists and antagonists are developed, they will allow the clearer definition of where modulation of muscarinic function will be of benefit in disease states.
