The capacity of presynaptic .alpha.-receptors on noradrenergic varicosities to modulate norepinephrine (NE) release in the rat neocortex was studied. Estimates of the affinity of 2 .alpha.-receptor agonists (NE and oxymetazoline) and 2 antagonists (phentolamine and yohimbine) for these receptors are presented. Cortex slices, previously labeled with -values of 8.0 and 7.4, respectively. From a comparison with previous data, presynaptic and postsynaptic .alpha.-receptors are apparently pharmacologically dissimilar.
