Cy alters the surface charge of human blood platelets as showed by biphasic changes in the electrophoretic mobility after addition of the compound. Low (non-aggregating) Cy concentrations increase platelet mobility whereas higher (aggregating) Cy solutions decrease mobility. Addition of Ca" inhibits the increase in mobility produced by low Cy concentrations. It appear.;, therefore, that the aggregating effect of Cy is in some way related to changes in the platelet membrane charges. Cy incubation in PRP inhibits collagen, thrombin, serotonin, the second waves of ADP and adrenaline-induced aggregation and the release of ADP and Pf4 produced by connective tissue. At variance with its inhibiting effects, Cy potentiates the first wave of ADP-induced aggregation. These findings, together with reports of other workers, suggest that the platelet release reaction is critically influenced by changes in the equilibrium between the positive and negative charges carried by platelet membranes.
