Elevation of peptidylproline hydroxylase activity and collagen synthesis in spontaneous primary mammary cancers of inbred mice

Peptidylproline hydroxylase (PPH) activity was located in the 15 000 X gn and 105 000 X gn supernatants of whole-tissue homogenates of spontaneous primary mammary cancer tissue and normal mammary gland in fat pad. Ascorbate, alpha -ketoglutarate and Fe2+ were required for enzyme activity of cancerous and normal tissues. PPH activity in mammary tissue from 5 high mammary cancer strains of inbred mice was 3-5 X higher than the level in normal mammary gland in fat pad of animals of comparable age. Activity was 2- to 3-fold greater in normal mammary gland in fat pads from 90-day-old mice compared with that in 300- to 500-day old mice. Hydroxyproline formation in cancerous tissue was 4 X greater than in normal mammary glands of animals of comparable age.
