A sensitive and accurate high performance liquid chromatography (HPLC) method has been used for the direct determination of allantoin in comfrey root. The allantoin in powdered comfrey root samples was completely extracted with 70% methanol using a soxhlet continuous extraction apparatus. Quantitative HPLC analysis of the extract was carried out using external and internal standardisation procedures. The results obtained from the two methods of analysis were in close agreement and indicated a much higher allantoin content for comfrey root than had been reported using other procedures.
