The direct effects of lhrh and three lhrh analogs on progesterone secretion by early pregnant human luteal cells in vitro

In animal experiments, it was found that luteinizing hormone releasing hormone (LHRH) and its analog (LHRH-A) have a direct inhibitory effect on ovarian progesterone production. However, this effect shows obvious species variation. Unlike the results from animal research, many data have indicated that LHRH and LHRH-A had no direct effect on cultured women luteal cells from normal menstrual cycle. But till now, little is known about the direct action of LHRH or LHRH-A on eary pregnant human luteal cells which, we consider, may be important in explanation of their antireproductive action. For this purpose, LHRH and three LHRH analogs with relatively high efficacy in inhibiting progesterone secretion by cultured luteal cells from pseudopregnant rats were used to observe their effect on progesterone seretion by early pregnant human luteal cells in vitro. Results showed that LHRH and LHRH-A have a direct but weak inhibitory effect on progesterone secretion by early pregnant human luteal cells. The maxium inhibition rate was 19.19% (P = 0.05). This inhibitory action was influenced by human chorionic gonadotropin(hCG) levels. The inhibitory action was comparatively obvious when added hCG concentration was low (hCG = 1 IU/ml), but was greatly decreased in the presence of high hCG concentration (hCG = 50 IU/ml).
