A modified ELISA technique for detecting antibodies against swine fever virus (SFV) in swine herds is described using lyophilized vaccine virus (Chinese strain of lactating rabbit lapinized SFV) as antigen and horseradish peroxidase-staphylococcal protein A (PPA) as conjugate. The results were compared with the serum neutralization test (SNT) in rabbits using 73 serum samples from pigs. Total agreement was found between ELISA and SNT in 68 serum samples and the sensitivity and specificity of the ELISA were 100% and 92.2%, respectively. The method was employed to detect SFV antibodies in 1305 serum samples collected from 129 piglets derived from 10 immune sows at different days after parturition. In piglets with maternal antibodies vaccinated at day 7 after parturition only a very slight increase in antibody levels occurred, beginning with the 20th day. For satisfactory vaccination of piglets it is necessary to determine the level of maternal antibodies before vaccination. The ELISA technique described is suitable for that purpose. Screening large number of serum samples is possible within a short time.
