Quantitative analysis of T and B lymphocytes and their subpopulations of irradiated mouse by scanning microspectrophotometry

On the basis of enzyme cytochemical staining, T and B lymphocytes and their subpopulations from lymph node suspensions of mouse were analyzed after 0, 1, 5 and 10 Gy .gamma.-irradiation with scanning microspectrophotometer. Results showed that the frequency profiles of T cells of normal mouse displayed a dense distribution, while that of B cells a disperse distribution. There were 5 subsets with different slopes in the frequency profiles of T and B cells. The characteristic parameters of the 4th subset of T cells approximated to those of T-helper cells (TH) identified cytochemically by dipeptidylaminopeptidase IV; and after 1, 5 and 10 Gy .gamma.-irradiation, the 4th subset of T cells corresponded with TH cells in radioresistance. It is suggested that both belonged to the same subpopulation of T lymphocytes in nature. The subsets of T and B lymphocytes had different radiosensitivity; B lymphocytes were more radiosensitive than T lymphocytes.
