Modified micro complement fixation test for detection of fowl pox virus antibodies

Micro complement fixation test was modified using 6% normal chicken serum to 1 unit of guinea pig complement for detection of complement fixing (CF) antibodies against fowl pox virus in vaccinated birds. The test was found specific and the CF antibodies could be detected even at 7 days post vaccination by intramuscular and feather follicle routes. However, CF antibody titer in 60% birds could not be detected up to 14 days post vaccination in the birds receiving the vaccines by oral and intranasal routes of immunization. Birds of different age groups immunized with 5 different fowl pox virus vaccines showed that all vaccines gave rise to low level of CF titer and maximum CF titer 1:16 was found with HP1 vaccine by intramuscular routine.
