Consistent and informative results regarding differentiation of sub-types and strains of foot-and-mouth disease (FMD) virus type O were obtained from complex and combined use of the plaque neutralization test, polyacrylamide gel electrophoresis (PAGE), and isoelectric focussing (IF). Selection of virus strains for vaccine production was made using the plaque neutralization test with antiserum collected from cattle and pigs that had been vaccinated once. The same test was used with good results in characterization of FMD strains and for detection of field virus variations in the course of an outbreak, when sera were used from vaccinated cattle, swine and guinea pig. PAGE and IF were suitable for characterization of field virus strains, determination of their relationships with other known strains, monitoring of virus strain properties in the course of an outbreak and characterization as well as follow-up of vaccine production strains.
