Effect of some beta-adrenoceptor antagonists on cerebrocortical acetylcholine release in rats

Propranolol, acebutolol, pindolol and butoxamine the anticonvulsant adrenergic .beta.-receptor antagonists were tested for their action on central cholinergic mechanism. The spontaneous release of acetylcholine (ACh) was reduced in the cerebral cortex of rats treated with 10-20 mg/kg of acebutolol and propranolol which stabilizes neuronal membrane at these dose levels. Pindolol and butoxamine which lack membrane stabilizing property, at these dose levels failed to show this effect. These results suggest that membrane stabilization and inhibition of ACh release are coincidental. It is proposed tha inhibition of cholinergic activity contributes to the anticonvulsant effect of high doses of .beta.-adrenoceptor antagonists that show membrane stabilizing property, because of the evidence that cerebrocortical release of ACh was diminished in rats treated with the anticonvulsants, phenobarbitone and diazepam.
