Effect of injection route and adjuvant ratio in antigen emulsion on antibody production

Using protein antigens of CRP and .alpha.fetoprotein, I examined the conditions for the production of the serum antibody. Various ratios of antigen emulsion with the Freund's adjuvant were injected into rabbits subcutaneously (sc), intracutaneously, intrafootpad or intraperitoneally (ip). High titers of serum antibody were produced by rabbits injected ip. In ip injections, the emulsion of antigen with the Freund's adjuvant in a 1:9 ratio (ip (9)) was more effective than that in the 1:1 ratio (ip (1)). Of three time immunizations, the 1st ip (9) injection determines the high productivity of serum antibody, even if the 2nd and 3rd injection routes were sc.
