Localization of fibronectin and actin in cultured rabbit keratocytes

To understand the relationship between fibronectin and activated keratocytes which appeared at the injured corneal stroma, we cultured rabbit corneal keratocytes and examined the dynamic movements of the cells by time-lapse cinematography and examined the changes in the localization of fibronectin and actin using double staining technique of immunofluorescent microscopy. Cultured keratocytes attached to substratum with round globular shape and then started to spread with many processes. At the early stage of the cultivation, fibronectin could be observed inside the cells. Then, fibronectin was observed outside the cells forming a part of extracellular matrix. When keratocytes spread, actin was observed as a stress fiber. At the edge of cellular processes, a close interaction between fibronectin and actin was observed. The present results suggested that fibronectin played an important role in the attachment of keratocytes to substratum which induced re-organization of intracellular actin filaments and promoted the migration of keratocytes.
